5株鸡传染性支气管炎病毒M基因克隆及序列分析

摘  要：
利用RT-PCR技术成功扩增出5株鸡传染性支气管炎病毒(IBV)分离株M基因全长cDNA,并将其克隆到pGEM-T Easy载体上,获得M基因的重组质粒。序列分析表明,5株IBV分离株间的M基因核苷酸同源性为89.0%～100.0%,与参考毒株核苷酸同源性为85.7%～99.3%;在M蛋白遗传进化树中,5株IBV分离株分居3个群;分离毒株M蛋白的α-螺旋结构主要分布在N端前100个氨基酸残基的肽段区,其他的部分主要是β-折叠和无规则卷曲结构,表明IBV分离株M蛋白在基因序列和二级结构上相对保守。

摘  要：
The entire cDNA of the M gene of five infectious bronchitis viruses(IBVs) were amplified by means of RT-PCR.They were cloned into pGEM-T Easy vectors.The nucleotide sequence analysis showed 89.0%-100% homology among the 5 isolates,and 85.7%-99.3% homology as compared to the reference control.Based on the phylogenetic analysis of the amino acids,these 5 isolates were in 3 separate groups.The α-helix regions were found mainly in the first 100 amino acids of the IBVs′ M proteins.The remainder was β-sheets and coils.Thus,the primary and secondary structures of the IBVs′ M gene appeared to be relatively conservative.
