乙酰胆碱酯酶固定化条件的研究

摘  要:
以戊二醛为交联剂固定乙酰胆碱酯酶,考察缓冲液浓度、pH值对乙酰胆碱酯酶固定化的影响,用正交实验考察牛血清白蛋白和戊二醛的浓度、交互作用及3种固定化膜材料对固定化效率的影响。确定固定化载体壳聚糖膜、0.01%戊二醛、2%牛血清白蛋白、pH值6.0、0.05mol/L磷酸缓冲液为固定化优化条件,当抑制率标准采用50%时,该方法得到的固定化乙酰胆碱酯酶膜对敌敌畏的检测浓度达0.53μg/L。

摘  要:
Acetylcholinesterase was immobilized with glutaraldehyde as a cross-linking agent, the concentration and pH value of phosphate buffer which affect immobilization, were explored.The concentration of bovine serum albumin and glutaralde-hyde, as well as, their interaction and three types of membranes, which affected immobilization, were analyzed by orthogonal matrix method.The results showed that the optimal condition was:chitosan membrane, 0.01% glutaraldehyde, 2% BSA, 0.05 mol/L phosphate buffer(pH 6.0).Acetylcholinesterase membrane prepared under the optimal condition, and the standard inhibition was 50%,the detection limit to dichlorvos reached 0.53 μg/mL.
