虫草发酵菌丝体中有效成分麦角甾醇的检测

摘  要:
为了建立虫草菌丝体中麦角甾醇的快速定量分析方法。以戴氏虫草、江西虫草的发酵菌丝体为研究材料,采用超声波法提取发酵菌丝体中游离麦角甾醇,利用HPLC-DAD法定量分析麦角甾醇。结果表明,麦角甾醇在0.46~370.00μg/mL有良好的线性关系(R2=0.9979),其检测限和定量限分别为3.50×10-3μg/mL和3.5×10-2μg/mL,灵敏度较高。该方法稳定性、重复性、精密度和准确度良好,其相对标准偏差均在3%以内,尚在HPLC检测允许范围内。戴氏虫草、江西虫草发酵菌丝体中游离麦角甾醇的含量分别为0.165mg/g和0.736mg/g。

摘  要:
Ergosterol,a major bioactive ingredient of fermented mycelia in C.taii and C.jiangxiensis was extracted by ultrasonic extraction,then a rapid and effective method HPLC-DAD was developed for the quantitative analysis in this study.The calibration curve showed good linearity(R2=0.9979)in the range of 0.46~370.00μg/mL,and the limit of detection(LOD)and limit of quantification(LOQ)were 3.50×10-3 μg/mL and 3.5×10-2 μg/mL,respectively.The method was simple,rapid,and accurate,and with high precision,stability,reproducibility for quantitative determination of ergosterol in fermented mycelia of Cordyceps with less than 3% of relative standard deviation.The content of free-ergosterol was determined in fermented mycelia from two Cordyceps species including C.jiangxiensis and C.taii with this method,and the corresponding content were 0.736and 0.165mg/g,respectively.
