单糖对猪精液碱性N-乙酰-β-D-氨基葡萄糖苷酶的影响

摘  要:
以D-果糖、葡萄糖、D-甘露糖和N-乙酰-D-氨基葡萄糖(NAG)为效应物,研究它们对猪精液碱性N-乙酰-β-D-氨基葡萄糖苷酶(NAGase,EC3.3.1.52)活力的影响及抑制机理.结果表明:在特定浓度范围内,低浓度的葡萄糖对NAGase活力表现为激活作用,而D-果糖、D-甘露糖和NAG对酶活力均表现为抑制作用,抑制程度从高到低依次为D-甘露糖、NAG、D-果糖;D-甘露糖表现为可逆的竞争型抑制,抑制常数KI为18.36 mmol·L-1;NAG表现为可逆的混合型抑制,抑制常数KI和KIS分别为16.98和55.26 mmol·L-1.

摘  要:
The inhibitory effect and mechanism of fructose,glucose,mannose and N-acetyl-D-glucosamine(NAG) on the activitv of the alkaline N-acetyl-β-D-glucosaminidase(NAGase,EC3.3.1.52) from procine semen have been studied.The results showed that lower concentrations glucose activated the enzyme,while fructose,mannose and NAG inhibited the enzyme in certain concentrations.The degree of inhibition was mannose>NAG>fructose.Mannose was reversible competitive inhibitor,with inhibition constants of free enzyme(KI) was 18.36 mmol·L-1.NAG was a mixed-type inhibitor.The inhibition constants of free enzyme(KI) and enzyme-substrate complex(KIS) with NAG were determined to be 16.98 and 55.26 mmol·L-1,respectively.
