温泉中降解纤维素嗜热细菌的分离与鉴定

摘  要:
采用纯培养的方法,从美国内达华州温泉和中国福建永泰温泉分离得到27株嗜热细菌,从中筛选得到产纤维素酶的嗜热细菌并进行了16S rDNA鉴定。结果表明,LY7和LY8是产纤维素酶的嗜热细菌,菌株LY7与脂环酸芽孢杆菌属(Alicyclobacillus)的同源性达到99%,菌株LY8与土芽孢杆菌属(Geobacillus)的同源性达到99%,生长温度范围在40~70°C之间,最适温度65°C。酶学性质分析表明,LY8的内切酶酶活高达145 IU/mL。

摘  要:
Culture-based approach was used to isolate thermophiles from hot spring. In total, 27 thermophilic bacterial strains were isolated from hot springs in Nevada of USA and Yongtai hot spring in Fujian province of China. Superior cellulose and hemicellulose decomposing strains were screened and identified by 16S rDNA. The results showed that LY7 and LY8 degrading cellulose were indentified as Alicyclobacillus sp. LY7 and Geobacillus sp. LY8. Geobacillus sp. LY8 could be cultured at temperature ranging from 40 ℃ to 70 ℃, with the optimal temperature of 65 ℃. The β-glucosidase activity of LY8 was 145IU / mL.
