双峰驼排卵机制的研究Ⅰ.促黄体素(LH)的放射免疫测定技术

本实验以羊参考制剂 NIH—LH—S_(18)为标准品,利用抗羊 LH 血清—放射性碘标记羊 LH 异源放射免疫测定系统(双抗体法)测定了双峰母驼卵泡发育期和精液诱导排卵前后血液中 LH 的水平。对此测定方法的特异性、灵敏度、准确度和精确度均分别进行了相应的检验,证明这一异源放射免疫测定系统可以对双峰驼血液中的 LH 进行测定,为双峰驼排卵机制的研究提供了一种有用的手段。测定时标准曲线的范围为0.125~32.0ng,灵敏度为0.1ng。一次测定内的变异系数为5.84%,测定间的变异系数为17.38%。应用此技术测得两头母驼的4个卵泡发育期中血液内 LH 的基础水平为2.70±1.29ng/ml;两例母驼在人工输精后5~8小时内血液中 LH 的峰值为11.20±4.67ng/ml,约为基础水平的4倍;经直肠检查,在 LH 峰值后40小时左右排卵。

摘  要:
A double antibody method of radioimmunoassay for measuring serum or plasma luteinizing hormone in bactrian camel has been developed by using rabbit anti-ovine LH serum and radioiodinated ovine LH.In this heterogeneous system the dose-response curve of ovine LH was paralled to that of the dilution curve of camel plasma.The specificity,accu- racy,precision and sensitivity of this assay were examined.As it is sh- own in this study,this assay system is demonstrated to be suitable for measuring the serum and plasma LH of camels.The ovine NIH-LH-S_(18) preparation was used as the standard and the sensitivity of this assay was found to be 0.1ng per tube.The coefficients of variation within a single assay and between assays were 5.84% and 17.38% respectively. It was found that during the follicular phase of two experimental camels,the basic level of serum or plasma LH was 2.70±29ng]ml.A peak value of 11.20±4.67ng/ml was found 5-8 hours after artificial in- semination and the ovulation occured about 40 hours following the ap- pearance of the LH peak.
