藏红花素对冻融牛精子获能参数、抗氧化及抗凋亡的影响

摘  要:
本研究的主要目的是评估藏红花素(crocin)对冻融牛精子获能参数、抗氧化及抗凋亡的影响。试验分对照组和藏红花素处理组(浓度分别为0.5、1、1.5和2 mmol/L),样本冻融处理后分别检测了获能参数、抗氧化相关基因及抗凋亡相关基因mRNA的表达。结果显示:1)0.5 mmol/L处理组冻融牛精子质量较佳,与对照组无显著差异而与其他处理组相比差异显著;(2)0.5 mmol/L处理组冻融牛精子获能处理后蛋白磷酸化水平显著高于其他处组(P<0.05);3)0.5 mmol/L处理组冻融牛精子抗氧化相关基因CAT与GPX基因mRNA的表达量显著高于其它组(P<0.05);4)0.5 mmol/L处理组冻融牛精子凋亡相关基因Caspase-3,TNF-α基因mRNA表达量显著低于其他组(P<0.05)。结论:牛精子冻融前藏红花素处理显著改善冻融牛精子获能参数、抗氧化及抗凋亡能力,添加量0.5 mmol/L藏红花素效果最佳。

摘  要:
The main objective of this study was to evaluate the effects of crocin on capacitation parameters, antioxidant and apoptosis of frozen-thawed bovine sperm. The experiment was divided into control group and crocin treatment group(concentrations of 0.5, 1, 1.5 and 2 mmol/L, respectively). The capacitation parameters, mRNA expression of antioxidant related genes and mRNA expression of anti-apoptotic related genes were detected after frozenthawed treatment. The results showed that frozen-thaw bovine sperm quality was better in the 0.5 mmol/L treatment group, there was no significant difference with the control group and significant difference with other treatment groups. The protein phosphorylation level of frozen-thaw bovine sperm treated with capacitation was significantly higher in the 0.5 mmol/L treatment group than in other groups(P<0.05). mRNA expression levels of CAT and GPX genes related to anti-oxidation in frozen-thaw bovine sperm in the 0.5 mmol/L treatment group were significantly higher than those in other groups(P<0.05). mRNA expression levels of apoptosis-related genes Caspase-3 and TNF-α genes in frozen-thaw bovine sperm treated with 0.5 mmol/L were significantly lower than those in other groups(P<0.05). In conclusion, crocin treatment before frozen-thaw significantly improved the capacitation parameters, antioxidant and anti-apoptosis ability of frozen-thawed bovine sperm. The effect of adding 0.5 mmol/L crocin was the best.
