摘  要:
以松茸为试材,使用超高效液相色谱-三重四级杆串联质谱法(ultrahigh-performance LC and tandem mass spectrometry,UHPLC-MS/MS)快速测定麦角甾醇含量,为松茸中麦角甾醇的进一步研究提供技术支持.采用Waters ACQUITY UPLC BEH C18(2.1 mm×100 mm,1.7 μm)色谱柱分离,甲醇等度洗脱,流速 0.3 mL·min-1,柱温 30℃,进样量 5 μL,串联质谱为大气压化学电离(APCI)源正离子模式、选择反应监测(SRM)模式.结果表明,麦角甾醇在 50~1 000 ng·mL-1 线性关系良好,相关系数大于 0.999;平均回收率为 92.1%、90.1%、98.5%,相对标准偏差为 2.49%、1.57%、1.99%.该方法具有检测时间短、灵敏性高的优点,可以满足松茸中麦角甾醇的批量检测.

摘  要:
Using Tricholoma matsutake as the test material,the content of ergosterol was rapidly determined by ultrahigh-per-formance LC and tandem mass spectrometry method,which can provide technical support for the further study of ergosterol in T.matsutake.The separation was performed on a Waters ACQUITY UPLC BEH C18(2.1 mm×100 mm,1.7 μm)column with iso-cratic elution methanol at a flow rate of 0.3 mL·min-1,column temperature of 30℃,injection volume of 5 μL,and tandem mass spectrometry was atmospheric pressure chemical ionization(APCI)source in positive ion mode and selected reaction monitoring(SRM)mode.The results showed that ergosterol had a good linear relationship in the range of 50-1 000 ng·mL-1,and the corre-lation coefficient was greater than 0.999.The average recoveries were 92.1%,90.1%and 98.5%with the relative standard devi-ations of 2.49%,1.57%and 1.99%.This method has the advantages of short detection time and high sensitivity,and can meet the batch detection of ergosterol in T.matsutake.
