摘  要:
【目的】通过分析KAP6.1基因的多态性,研究KAP6.1基因与绵羊羊毛性状的相关性,为进一步研究KAP6.1基因功能及细毛羊分子育种提供基础。【方法】以693只中国美利奴羊(新疆军垦型)为实验材料,采用PCR技术、SSCP技术和克隆测序的方法分析KAP6.1基因的多态性,并开展与毛性状的关联性分析。【结果】KAP6.1基因存在C159T碱基突变,经最小二乘拟合线性模型分析,C159T位点AA、BB和AB基因型间的平均毛纤维直径、卷曲度、毛长、净毛率和密度差异不显著(P>0.05),而BB基因型平均污毛产量显著高于AA和AB基因型(P<0.05)。【结论】KAP6.1基因可作为绵羊污毛产量的候选基因之一,BB基因型可作为分子标记用于选择污毛产量高的个体。

摘  要:
【Objective】The Purpose of this study was to determine the association between KAP6.1 gene and the wool traits by polymorphism and association analysis,and provide a foundation for further studying the function of KAP 6.1 gene.【Method】PCR-SSCP gene cloning and sequencing technology were used to analyze polymorphisms of KAP6.1 gene in 693 Chinese Merino(Xinjiang Junken type).Correlation of wool traits and KAP 6.1 gene was analyzed by JMP 4.0 software.【Result】C→T base substitution existed in 159 sites of KAP6.1 gene.By least-squares fit linear model analysis,there was no significant association among AA,AB and BB genotypes of C159T site in average wool fiber diameter,wool curvature,wool fibre length, clean fleece rate and wool density,but the of BB genotype was significandy higher than AA and AB genotypes (P<0.05).【Conclusion】KAP6.1 gene can be regarded as a candidate gene of average wool fleece weight, and BB genotype can be used as molecular markers for selecting individual of high wool fleece weight.
