捕食线虫真菌的潮霉素抗性基因标记及其在土壤抑菌研究中的应用

摘  要:
用潮霉素磷酸转移酶基因(hygB)转化了捕食线虫真菌蠕虫节丛孢Arthrobotrysvermicola;转化率为1~3个转化子μg-1DNA;转化子的潮霉素抗性稳定性为87 5%。选取了一个潮霉素抗性稳定、生长和形态与原始菌株相似的转化子Tav36,研究了其在土壤中的生长及其对线虫的捕食,证明了此转化子能够在自然土壤中正常地生长和捕食线虫,但其生长速率和对线虫的致病能力受到了土壤抑菌作用的抑制。

摘  要:
We transformed the nematode-trapping fungus Arthrobotrys vermicola with hygromycin B phosphotransferase gene (hygB) using restriction enzyme-mediated integration (REMI).The transformation frequency was 1-3 transformants μg~(-1) DNA and the transformants stability of hygromycin B resistance was 87.5 %. A mitotic stable transformant Tav36, without obvious difference in vegetative growth and pathogenicity to nematodes compared with that of the wild type, was selected for the detection and monitoring fungal growth and pathogenicity activity in natural soil.Results proved that Tav36 could grow normally in natural soil and trap nematodes,however,the vegetative growth and the pathogenicity to nematodes of Tav36 in natural soil were restrained by fungistasis of soil.
