肠出血性大肠杆菌O157∶H7 LAMP-LFD快速检测方法的建立及初步应用

摘  要:
针对肠出血性大肠杆菌O157∶H7(EHEC O157∶H7)的rfbE基因保守序列设计特异引物,通过优化反应体系、反应温度和反应时间等参数,建立了一种快速、简便的肠出血性大肠杆菌O157∶H7的环介导等温扩增-横向流动试纸条(LAMP-LFD)检测方法。特异性试验结果显示,该方法与大肠杆菌O8∶K88和O141∶K99、无乳链球菌、停乳链球菌、金黄色葡萄球菌、痢疾志贺菌、产单核细胞李氏杆菌、表皮葡萄球菌、肠炎沙门菌、猪链球菌2型、副猪嗜血杆菌、胸膜肺炎放线杆菌等12种细菌均无交叉反应。灵敏度试验结果表明,该方法对纯培养的EHEC O157∶H7的检测下限为13CFU/mL,污染食品中EHEC O157∶H7的检测下限为18CFU/mL。研究结果表明,该方法特异性好,灵敏度高,操作简便,可肉眼直接判定结果,1h内完成检测,在食品卫生检验和临床疾病诊断方面具有良好的应用前景。

摘  要:
A set of six primers were designed specifically to recognize the highly conserved sequence of the rfbEgene of enterohemorrhagic Escherichia coli O157∶H7(EHEC O157∶H7).A rapid method for detection of EHEC O157∶H7was developed by loop-mediated isothermal amplification(LAMP)combined with a lateral flow dipstick technology(LFD),namely LAMP-LED.The results showed the LAMP-LED method can specifically detect the EHEC O157∶H7and no cross reaction with other 12 bacteria,including E.coli O8∶K88,E.coli O141∶K99,Streptococcus agalactiae,Streptococcus dysgalactiae,Staphylococcus aureus,Shigella dysenteriae,Listeria monocytogenes,Staphylococcus epidermidis,Salmonella enteritidis,Streptococcus suis type 2,Haemophilus parasuis and Actinobacillus pleuropneumoniae.The detection limit of the LAMP-LED method was 13CFU/mL for pure culture of EHEC O157∶H7,and 18CFU/mL for the EHEC O157∶H7pollution food samples of artificial simulation.
