摘  要:
为了研究不同浓度的肠出血性大肠杆菌(EHEC)O157∶H7 Intimin和Tir-Tccp蛋白质对接受1010 CFUEHEC O157∶H7小鼠的免疫保护作用,将Intimin和Tir-Tccp蛋白质和佐剂充分乳化后,通过皮下多点注射免疫小鼠,对照组注射等量的磷酸盐缓冲液。共免疫2次,间隔14 d,末次免疫后14 d攻毒1只小鼠EHEC O157∶H7 1010CFU。每次免疫后的14 d通过断尾采血,用间接ELISA法检测血清抗体。攻毒后,观察小鼠精神状态,记录死亡数并检测排菌量。ELISA检测结果表明,Tir-Tccp蛋白质诱导小鼠产生IgG抗体水平明显升高,而Intimin诱导产生的IgG抗体升高不明显。攻毒后,各免疫组小鼠均死亡1只,存活14只,存活率为93.3%;对照组小鼠死亡4只,存活6只,存活率为60.0%。免疫组在攻毒后6 d,粪便中检测不到EHEC O157∶H7,对照组在攻毒后13 d仍有较高的排菌量。表明Intimin和Tir-Tccp蛋白质对小鼠起到了有效的免疫保护作用,能够减少EHEC O157∶H7在小鼠体内的定殖。

摘  要:
The objective was to evaluate the protective efficacy against 1010 CFU of enterohaemorrhagic Escherichia coli(EHEC) O157∶H7 after subcutaneously immunized with intimin and Tir-Tccp recombinant protein at different concentrations.After emulsifying intimin and Tir-Tccp proteins with adjuvant,the Balb/c mice of experimental groups were subcutaneously immunized twice(on days 0 and 14) with the protein,while the Balb/c mice of control group were subcutaneously immunized twice with equivalent phosphate buffered saline(PBS).On the 14th day following the second immunization,all the mice were orally infected with 1010 CFU of EHEC O157∶H7.Serum samples were collected via cutting off the tail on day 14 after every immunization and IgG antibody was determined by ELISA.The mental states of all the mice were observed,and the mortality and shed bacteria quantity were monitored after challenge.The result of ELISA demonstrated that the Tir-Tccp protein elicited high titre antibody but the levels of antibody induced by intimin was low.After challenge,one mouse died in each of the immunized groups and the survival rate was 93.3%,while the survival rate of the control group was 60.0%.There was no EHEC O157∶H7 in the fecal samples of two immunized groups on the 6th day after challenged,but control group mice still shed high level of the organism on 13th day.The results demonstrated that Intimin and Tir-Tccp recombinant protein significantly reduced shedding of EHEC O157∶H7 from experimentally infected mice.
