平邑甜茶叶片不定芽再生及NO的效应

摘  要:
以平邑甜茶继代组培苗叶片为材料,研究了一氧化氮(NO)供体——硝普钠(SNP)、6-BA、NAA、2,4-D、ZT和光暗条件对平邑甜茶叶片不定芽再生的影响。结果表明,6-BA、ZT、NAA和暗培养均有利于叶片不定芽的分化;在含有生长素和细胞分裂素的MS培养基中,加入6.0和12.0mg·L-1的SNP可极显著诱导叶片不定芽的分化;以MS+6.0mg·L-1BA+0.5mg·L-1NAA+0.2mg·L-1ZT+6.0mg·L-1SNP为培养基,暗培养15d,叶片不定芽分化效果最好,再生率达67.8%。

摘  要:
The effects of nitric oxide donor-sodium nitroprusside(SNP),6-BA,NAA,2,4-D,ZT and dark treatment on adventitious shoot regeneration from leaves of Malus hupehensis Rehd.were studied.The results showed that 6-BA,ZT,NAA and dark treatment could increase adventitious shoot differentiation.In the presence of auxin and cytokinin,6.0 and 12.0 mg·L-1SNP could significantly induce leaf adventitious shoot differentiation.The best result was achieved in the medium MS+6.0 mg·L-1 BA +0.5 mg·L-1 NAA + 0.2 mg·L-1 ZT+ 6.0 mg·L-1 SNP,in which the percentage of adventitious shoot regeneration were 67.8% after dark treatment for 15 d.
