摘  要:
为探讨小麦中TabZIP1应答低温胁迫及ABA调控信号的响应机制,以强抗寒性冬小麦品种东农冬麦1号(DN1)和弱抗寒性品种济麦22(JM22)为材料,在大田自然降温条件下,分别于5℃、0℃、-10℃、-25℃取样叶片和分蘖节,通过RT-PCR分析TabZIP1的表达模式,于不同温度下探究ABA对DN1分蘖节TabZIP1表达模式的调控,并对其生物信息学进行分析。结果表明,TabZIP1全长1 167bp,编码388个氨基酸,其表达产物为疏水蛋白,定位于线粒体,无信号肽或跨膜结构域;TabZIP1蛋白二级结构为mixed型。DN1叶片和分蘖节中的TabZIP1表达量均在-10℃时达到最高,且分蘖节中的表达量高于叶片;而JM22分蘖节中的TabZIP1表达量在0℃时达到峰值,叶片中的表达量却在-10℃达到最高。综合分析,DN1分蘖节中的TabZIP1表达量明显高于JM22。ABA处理后,DN1分蘖节中TabZIP1的表达量随温度的降低呈明显升高趋势,在-25℃时达到峰值。可见,ABA正调控TabZIP1的表达,有助于提高冬小麦的抗寒性。

摘  要:
In order to investigate the response of TabZIP1 to chilling stress and ABA regulation signal response mechanism,with strong cold resistance winter wheat variety‘Dongnongdongmai 1'and weak cold resistance cultivar ‘Jimai 22'as experimental materials,samples were collected at 5 ℃,0 ℃,-10 ℃,-25 ℃,respectively,in the field of natural under cool conditions.The expression pattern of TabZIP1 in the blade and tiller node were analysed by RT-PCR.The regulation of ABA on TabZIP1 expression pattern of tillering in Dongnongdongmai 1under different temperatures was explored with bioinformatics analysis.The results showed that,the full length of TabZIP1 is 1 167 bp,encoding 388 amino acids,and the expression product is a hydrophobic protein,localized in the mitochondria,with no signal peptide or transmembrane domain;and the secondary structure of TabZIP1 protein is mixed type.The expression level of TabZIP1 in leaves and tiller node of Dongnongdongmai 1are both highest at-10 ℃,and higher in tiller node than in blade.While in Jimai 22,the expression level of TabZIP1reached the maximum at 0℃in tiller node,but at-10℃in blade.Based on comprehensive analysis,the expression level of TabZIP1 in tillering node of Dongnongdongmai 1is highest,which is significantly higher than that of Jimai 22.With ABA treatment,the expression level of TabZIP1 in tillering node of Dongnongdongmai 1was significantly reduced with temperature increasing,with a maximum at 25 ℃.It is suggested that ABA treatment positively regulated the expression of TabZIP1,thereby improving the cold resistance of winter wheat.
