摘  要:
为了研究人类动脉粥样硬化性疾病及其发病机制,采用高脂饲料饲喂合并球囊拉伤的方法,模拟人类自然发病过程,建立小型猪颈总动脉粥样硬化动物模型。实验结果显示:饲喂高脂饲料后第30天造模,动物存活率100%,随时间的增加体重逐渐升高;与基础值相比,血液中甘油三酯(TG)含量变化不显著,总胆固醇(TC)、高密度脂蛋白(HDL-C)和低密度脂蛋白(LDL-C)含量变化显著(P!0.05);饲喂高脂饲料后第120天,与基础值相比,总胆固醇含量升至其5.6倍,低密度脂蛋白含量升至其8.4倍,高密度脂蛋白含量升至其4.4倍;动脉粥样硬化指数(AI值)均大于4;病理学观察显示血管损伤部位管腔狭窄,病变符合动脉粥样硬化的基本特征。上述结果表明,在本实验条件下,可以在4个月内成功复制小型猪动脉粥样硬化(As)模型,与其他现有的As动物模型相比,具有成模时间短,方法简便易行的特点。

摘  要:
To study human atherosclerotic disease and its pathogenesis,natural pathogenesis of human beings was simulated and a minipig model of carotid atherosclerosis was established by high-fat feeding combined with balloon injury.The results demonstrated that at the 30 th day after high-fat feeding,the survival rate of the minipigs was 100%and the body weights gradually increased over time.Compared with basic levels,triglyceride(TG)content showed no significant change.However,total cholesterol(TC),highdensity lipoprotein cholesterol(HDL-C)and low-density lipoprotein cholesterol(LDL-C)contents changed evidently(P<0.05).At the 120 th day of high-fat feeding,the contents of TC,LDL-C and HDL-C was 5.6,8.4 and 4.4 times of the basic levels,respectively.All the atherogenic indexes(AI)were higher than 4.Pathological observation showed luminal stenosis in the site of vascular injury,and the pathological changes accorded with the basic characteristics of atherosclerosis.The above results indicated that under our experimental conditions,the minipig model of atherosclerosis(As)could be successfully copied within4 months,and was characterized by short time of establishment and handy operation as compared with other existing As animal models.
