鸡滑液囊支原体的RT-PCR检测及基因序列分析

摘  要:
为确诊某鸡场肉鸡发病死亡的病因,通过临床症状、病理变化、RT-PCR检测、病原S1基因序列分析进行综合诊断。结果:(1)病鸡表现食欲不振、精神沉郁、跛行及无法站立、关节肿大等症状,剖检发现肿大关节内有大量黏液。(2)鸡滑液囊支原体RT-PCR扩增出现207 bp特异性条带,与预期结果一致。(3)病原S1基因序列分析显示与GenBank公布的鸡滑液囊支原体基因序列同源性均达到99%,构建系统进化树分析显示与GenBank公布的参考株聚为1支。综合诊断为鸡滑液囊支原体感染。

摘  要:
In order to identify the cause of death in a chicken farm,the comprehensive diagnosis was made by clinical symptoms,pathological changes,RT-PCR detection,and pathogen S1 gene sequence analysis. Result:( 1) The sick chicken showed symptoms such as loss of appetite,depression,lameness and inability to stand,joint swelling,etc.,necropsy found a large amount of mucus in the enlarged joint.( 2) The 207 bp specific band appeared in the RT-PCR of Mycoplasma synoviae,which was consistent with the expected results.( 3) Sequence analysis of the pathogen S1 gene showed that the sequence homology with the Mycoplasma synoviae gene published by Gen Bank reached 99%. The phylogenetic tree analysis showed that the reference strain published by GenBank was clustered into one. The diagnosis was Mycoplasma synoviae infection.
