不同品种小麦成熟胚愈伤组织的诱导与分化

摘  要:
以4种不同基因型小麦(Triticum aestivum)品种成熟胚作为外植体进行培养,研究不同培养条件对愈伤组织诱导及植株分化再生的影响。结果显示不同小麦品种对培养条件的反应各不相同。各品种小麦成熟胚的最高愈伤诱导率所需的2,4-D浓度不同,石4185和科麦一号的最适浓度为2.0 mg/L;扬麦12和Bobwhite的最适浓度为4.0 mg/L。4种小麦品种成熟胚愈伤组织再分化的最适条件也有所差别,在MS培养基中添加2.0 mg/L KT和0.1 mg/L NAA利于石4185和Bobwhite成熟胚的愈伤组织分化;科麦一号成熟胚的分化最适条件为1.0 mg/L KT和0.5 mg/L NAA;扬麦12成熟胚的分化则采用1.0 mg/L KT和0.1 mg/L NAA效果最佳。此外,继代培养时间的延长会不同程度地影响小麦成熟胚愈伤的分化。

摘  要:
Mature embryos from four wheat varieties were cultured in different media to study the induction of the callus and plants regeneration. The results indicated that different varieties showed significantly different responses under various culture conditions. The most suitable concentration of 2,4-D for callus induction of mature embryo from four wheat varieties was different. The optimal 2,4-D concentration was 2.0 mg / L for Shi4185 and Kemai NO.1,and 4.0 mg / L for Yangmai 12 and Bobwhite,separately. The best condition for callus redifferentiation also varied. MS medium supplemented with 2.0 mg / L KT and0.1 mg / L NAA for Shi4185 and Bobwhite,1.0 mg / L KT and 0.5 mg / L NAA for Kemai NO.1,and 1.0 mg / L KT and 0.1 mg / L NAA for Yangmai 12 was conducive to improving differentiation and regeneration, respectively. In addition, with the extension of culture time,the differentiation rate of callus of wheat mature embryos would decline to different degree.
