山西运城盐湖嗜盐细菌的系统发育与种群多样性

摘  要:
为嗜盐微生物资源的保护、开发和利用提供理论依据,采用分子生物学方法对运城盐湖沉积水土样品的嗜盐细菌进行分离,并采用克隆文库法构建嗜盐菌可培养文库和未培养文库,对2个不同文库环境样品中细菌群落进行系统发育分析及其多样性、丰富度和优势度研究。结果表明:可培养文库的主要类群为厚壁菌门(Firmicutes,55%)和变形菌门(Proteobacteria,45%),未培养文库的主要类群为变形菌门(Proteobacteria,43%)、厚壁菌门(Firmicutes,28%)和拟杆菌门(Bacteroidetes,10%)等。可培养文库和未培养文库的Shannon多样性指数(H′)分别为2.63和4.83,二者的多样性虽都比较丰富,但未培养文库的多样性高于可培养文库,表明其物种更为丰富。

摘  要:
For the sake of providing theoretical basis for protection,development and utilization of the halophilic microorganism resources,using molecular biology methods in the clone library construction of halophilic bacteria cultivation library,and bacteria in environmental samples in two different libraries of phylogenetic analysis and the diversity of its abundance and dominance were studied.Results:Firmicutes(55%)and Proteobacteria(45%)were the major phylogeny groups for cultured library construction,and Proteobacteria(43%),Firmicutes(28%)and Bacteroidetes(10%)for uncultured library construction.Shannon index for cultured and uncultured library was respectively 2.63 and 4.83,both of which were abundant,while the latter was higher than the former,indicating the species was more abundant.
