藏系绵羊复胃差异表达基因的筛选及功能分析

摘  要:
为阐明藏绵羊瘤胃、网胃、瓣胃与皱胃功能差异的分子机理,试验以藏绵羊为研究对象,采用抑制消减杂交法筛选藏绵羊瘤胃、网胃、瓣胃与皱胃差异表达基因并进行生物信息学分析。结果表明:成功构建了6个藏绵羊瘤胃、网胃、瓣胃与皱胃差异表达的正反向消减c DNA文库;得到了皱胃特异表达的30个EST片段,瘤胃、网胃、瓣胃特异表达的10个EST片段。经Blast X对比发现5个EST片段与已知基因具有较高的序列相似性,且都在皱胃中特异表达。其中胃蛋白酶A、Napsin-A参与蛋白质水解;胃溶菌酶参与菌体蛋白的消化过程;胃饥饿激素调节食欲且促进相关消化酶分泌;锌指蛋白调节胃酸分泌。说明皱胃具有真正的消化功能,而瘤胃、网胃、瓣胃不能分泌消化酶。瘤胃能利用微生物将摄入的饲草发酵分解成小颗粒物质,网胃、瓣胃起筛滤作用。

摘  要:
To clarify the differences of molecular mechanism in Tibetan sheep's rumen,omasum,reticulum and abomasum function. Suppression subtractive hybridization( SSH) was used to screen the differentially expressed genes in compound stomach. Then,the obtained genes were analyzed by bioinformatics method. The results suggested that six forward and reverse subtracted c DNA libraries of abomasum and the rumen,reticulum,omasum were constructed successfully. And thirty expressed sequence tag( EST) fragments were obtained from the SSH c DNA library of abomasum and ten EST fragments were obtained from rumen,reticulum and omasum. Blast X analysis revealed that five EST fragments had highly similarity to the known proteins,and all of them were differentially expressed in the abomasum. Pepsin A,Napsin-A participated in protein hydrolysis. Gastric lysozyme were involved in the digestion of bacterial proteins. Ghrelin regulates appetitie and promotes secretion of digestive enzymes. Zinc finger protein regulates gastric acid secretion. This experiment demonstrated that abomasum was a truly functional digestive organs. While rumen,reticulum and omasum had no digestive enzyme secretion function. The forage was decomposed into small particles by microbial fermentation,then sieved by reticulum and omasum.
