白羽番鸭脂联素基因SNP与肌内脂肪和血清总胆固醇的关联分析

摘  要:
采用PCR-SSCP法对白羽番鸭脂联素基因全编码区和内含子进行多态性检测,并分析其多态对肌内脂肪(IMF)和血清总胆固醇(TC)的遗传效应,为番鸭分子标记辅助选择提供参考。结果表明:3个SNP分别在外显子1和外显子2的编码区发生A167G和G711A突变,为沉默突变,在内含子发生C290T突变;3个SNP位点均处于Hardy-Weinberg平衡状态(P>0.05),均表现为中度多态;关联分析表明,在A167G位点中,AA基因型个体的IMF和TC显著高于BB基因型个体(P<0.05);在C290T位点中,CC基因型个体的IMF显著高于TT基因型个体(P<0.05);其他各位点的各基因型与各性状间没有达到显著性差异(P>0.05)。该研究结果揭示,A167G和C290T位点对脂肪沉积可能有显著性的遗传效应,可作为番鸭的分子育种标记。

摘  要:
The whole coding region and intron of adiponectin gene were amplified with 4 pairs of primers designed from Cairina moschata DNA sequence(GQ873067) and its polymorphism was detected by PCR-SSCP method,and then its association with intramuscular fat(IMF) and serum total cholesterol(TC) were analysed.Three SNPs(A167G,T290C and G711A) were found.A167G and G711A,which belonged to silence mutations occurred in exon1 and exon2 coding region respectively,and T290C mutation was produced in intron.The three SNP loci presented medium polymorphism and were in accordance with Hardy-Weinberg equilibrium(P>0.05).Association analysis showed that the birds of genotype AA had significantly higher than genotype BB for IMF and TC in A167G locus(P<0.05).Then,the birds of genotype CC had significantly higher than genotype TT for IMF in T290C locus(P<0.05).The research suggested that the A167G and T290C loci had significant genetic effect on fatty deposition and may be used as markers of molecular breeding in White Muscovy.
