摘 要:
以大肠杆菌的6个管家基因(recA,aspC,ldh,gapdh,mdh,16S rRNA)作为候选的内参基因,大肠杆菌在不同浓度猪β防御素2(0、37.5、75、150 mg·L-1)的胁迫下培养不同时间(1 h和4 h)后,应用实时荧光定量PCR(RT-qPCR)技术比较这6个内参基因表达的稳定性,并通过GeNorm软件分析。结果表明,大肠杆菌中recA基因表达最稳定,其次是aspC和gapdh。并且16S rRNA的相对表达量最高,而ldh的表达量最低。

摘 要:
In this study,six housekeeping genes of E. coli( recA,aspC,ldh,gapdh,mdh,and 16 S rRNA) were selected as candidate reference genes,and the stability of the selected genes were identified by real-time quantitative PCR(RT-q PCR) and analyzed by Ge Norm software,after Escherichia coli were co-cultured with different concentrations of porcine beta defensin 2(0,37. 5,75 and 150 mg· L- 1) for different times(1 h and 4 h). The results showed that rec A gene had the most stable expression among the six housekeeper genes of Escherichia coli,asp C and gapdh followed. In addition,the relative expression of 16 S rRNA gene was the highest among the six housekeeper genes in Escherichia coli,and ldh was the lowest.
