摘  要:
采用微生物分离纯化技术,自江苏省连云港市某水产养殖公司采集的泥鳅样品病灶部位,分离到1株腐皮病病原细菌D5-7,经回归感染得以确认。通过菌落和细胞形态观察、生理生化试验及16S rRNA基因序列分析,将该菌株鉴定为杀鲑气单胞菌杀鲑亚种。以杀鲑气单胞菌D5-7为病原指示菌,采用滤纸片抑菌圈法试验,通过初筛和复筛,自泥鳅养殖池水样中分离筛选到菌株X8,对杀鲑气单胞菌D5-7具有较强的抑菌作用,抑菌圈直径达(14.2±0.10) mm,经鉴定菌株X8为解淀粉芽孢杆菌,适应温度20~40℃,盐度0~50。解淀粉芽孢杆菌X8对引起水产病害的其他病原如维氏气单胞菌、温和气单胞菌、嗜水气单胞菌、迟缓爱德华菌、哈维弧菌、鳗弧菌具有较强的抑菌作用,抑菌圈直径分别为(14.7±0.10) mm、(16.8±0.15) mm、(11.3±0.20) mm、(8.2±0.05) mm、(11.8±0.10) mm、(8.2±0.08) mm。在解淀粉芽孢杆菌X8的泥鳅养殖应用试验中,试验组比对照组体长增加1.40 cm,体质量增加2.44 g,酸性磷酸酶提高0.18 U/mL,试验组的这3项指标均与对照组差异极显著(P<0.01)。该研究结果可为水产病害的生物防治提供理论依据,有望研制成防治水产病害的生物制剂。

摘  要:
The pathogen D5-7 was isolated from the lesions of the loach Paramisgurnus dabryanus infected with putrid-skin disease collected from a breeding farm in Lianyungang, Jiangsu province by microbiological isolation technology, and confirmed by the recurrent infection. The strain was identified as Aerommads salmonicida ssp. salmonicida D5-7 based on the colonies and cell morphology observation, physiological and biochemical experiments, and 16 S rRNA gene sequence analysis. The further using D5-7 as an indicator bacterium, X8, was isolated from the water sample of the loach aquaculture ponds by the filter paper method. In the primary screen and double screen experiments, the strain X8 presented a strong antimicrobial activity against pathogen D5-7, with antibacterial circle diameter of(14.2±0.10) mm. The X8 strain was identified as Bacillus amyloliquefaciens based on the results by above methods same as D5-7 which was adapted to temperature of 20—40 ℃ and salinity of 0—50. It also had strong inhibitory against the other pathogens causing putrid-skin disease such as A. veronii, A. sobria, A. hydrophila, Edwardsiella tarda, V. harveyi and V. anguillarum, with antibacterial circle diameter of(14.7±0.10) mm,(16.8±0.15) mm,(11.3±0.20) mm,(8.2±0.05) mm,(11.8±0.10) mm, and(8.2±0.08) mm, respectively. In culture application experiment of X8, the loach in experimental group was had 1.40 cm longer in body length than the loach in control group did, the weight increased by 2.44 g, and the acid phosphatase(ACP) increased by 0.18(U/mL), in the three indicators between the experimental and the control groups were with very significant difference(P<0.01). The findings provide a theoretical basis for biological control of aquatic diseases, which can be developed as biological agents for the control of aquatic diseases.
