苇蘑液体培养条件的优化

摘  要:
通过苇蘑菌丝体液体培养的单因素试验,初步确定苇蘑的较适生长条件:pH值为5~7;温度25℃;转速100~200 r·min~(-1);装液量50~90 m L。再经正交试验对培养条件进行优化,得出苇蘑的最适制种工艺为:初始pH值为6;温度恒定为25℃;转速保持150 r·min~(-1);装液量为90 m L/250 m L。在该培养条件下,苇蘑的菌丝干质量可达到19.4 g·L~(-1)。在通气量为4.5 L·min~(-1)条件下进行苇蘑液体菌种的5 L发酵罐培养,在44 h即达峰值19.9 g·L~(-1)。

摘  要:
The optimum growth condition of liquid culture for Weimo was studied by the way of single factor test, including pH, temperature, rotating speed and liquid volume. The results showed that the range of growth conditions of Weimo are as follows: pH 5 ~7,temperature 25 ℃, rotating speed 100 ~200 r·min~(-1)and liquid volume 50 ~90 m L. Then the optimal spawn-making for Weimo was concluded by three factors three levels orthogonal experiment, and they are pH 6, temperature 25 ℃, rotating speed 150 r·min~(-1)and liquid volume 90 m L. Under the optimal growth condition, the dry weight of Weimo could reach 19.4 g·L~(-1). And Weimo spawns grew to a peak value 19.9 g·L~(-1)after cultured by 5 L fermenter, 4.5 L·min~(-1)ventilation, 44 h.
