摘  要:
为探究乳酸乳球菌Z-2(Lactococcus lactis Z-2)不同处理方式对鲤的益生作用,试验共设置L. lactis Z-2菌株6种不同处理[活菌及上清液的混合物(mixture of live cells and supernatant,LCS)、无细菌培养上清液(cell-free culture supernatant,CS)、活菌(live cells,LC)、菌热灭活(heat-killed cells,HK)、菌超声破碎(ultrasonic broken of cells,UB)和胞外多糖(exopolysaccharides of CS, EPS-2)]与鲤[体重(47.66±0.43)g]头肾单核细胞体外共培养,检测其对鲤头肾单核细胞的增殖能力和吞噬活性,及孵育液中一氧化氮(NO)和免疫相关细胞因子表达量的影响。结果表明:LCS、LC、UB和EPS-2处理组较之对照组,均能显著增强头肾细胞的增殖能力和吞噬活性,HK处理组仅能显著增强头肾细胞的吞噬活性(P<0.05),其中LC(1.580±0.032)和LCS(1.520±0.058)分别对鲤头肾细胞增殖能力和吞噬活性具有最积极的促进作用。除CS组外,其余处理组均能显著提高孵育液中NO的合成量(P<0.05),EPS-2[(1.960±0.043)μmol/L]效果最为显著。所有处理组均能显著增加头肾细胞中细胞因子TNF-α、IL-1β、IL-6和TGF-β的合成量(P<0.05),并且分别是LCS[(23.11±1.31)pg/mL]、HK[(991.82±55.86)pg/mL]、LCS[(147.98±8.25)pg/mL]/CS[(147.38±7.94)pg/mL]和LCS[(139.56±9.69)pg/mL]处理组具有最佳的促进效果;而仅有LCS、CS和EPS-2处理组能显著提升IL-10的合成量(P<0.05),且LCS[(81.82±3.03)pg/mL]具有最佳的促进效果。研究表明,L. lactis Z-2的不同处理方式均对鲤表现出一定的免疫调节作用。

摘  要:
To investigate the effects of the oral administration of different treatments for(mixture of live cells and supernatant, LCS; cell-free culture supernatant, CS; live cells, LC; heat-killed cells, HK, ultrasonic broken of cells, UB; exopolysaccharides of CS, EPS-2) L. lactis Z-2 on the probiotic performance in head kidney cells of Cyprinus carpio [(47.66±0.43) g], these different treatments of L. lactis Z-2 were co-cultured with the head kidney cells of the Cyprinus carpio. The proliferation and phagocytosis activition of nitric oxide(NO) and cytokines erpressionin the culture medium were determined. The re-sults showed that the preparation methed LCS,LC, UB and EPS-2 of L. lactis Z-2 significantlyenhanced the proliferation and phagocytic activity of the head kidney cells, HK treatment group only significantly enhanced the phagocytic activity of thehead kidney cells(P<0.05), LC(1.580±0.032) and LCS(1.520±0.058) had the most positive effect on theproliferation and phagocytic activity of head kidney cells, respectively. Except for CS group, the other treat-ment groups significantly increased the amount of NO synthesis in the incubation solution(P<0.05), the NOsynthesis effect of the EPS-2 [(1.960±0.043) μmol/L] was the most significant. The synthesis of cytokines(TNF-α, IL-1β, IL-6 and TGF-β) in head kidney cells was significantly increased in all treatment groups(P<0.05), and the LCS [(23.11±1.31) pg/mL], HK [(991.82±55.86) pg/mL], LCS [(147.98±8.25) pg/mL]/CS[(147.38±7.94) pg/mL] and LCS [(139.56±9.69) pg/mL] treatment groups had the best promotion effect re-spectively. Only the LCS, CS and EPS-2 treatment groups could significantly increase the synthesis ofIL-10(P<0.05), and the LCS [(81.82±3.03) pg/mL] had the best promotion effect on IL-10. Researchshowd that, the different treatment methods of L. Lactis Z-2 all showed a certain immunomodulatory ef-fect on common carp, which provided a theoretical basis for the rational use of potential probiotics and anew solution for the application of lactic acid bacteria to microecological agents.
