摘  要:
针对亚洲区域内O/PanasiaⅡ和A/Iran05口蹄疫流行毒给我国养殖业造成的严重威胁,以O/HN/93和A/GDMM/2013感染性克隆为骨架,替换了完整的结构蛋白编码基因P1,构建嵌合病毒作为疫苗储备毒株。分别用间接免疫荧光、RT-PCR、基因测序、电子显微镜、蚀斑表型及一步生长曲线鉴定和分析重组病毒。结果显示,成功拯救到含O/PanasiaⅡ和A/Iran05型毒株P1基因的嵌合口蹄疫病毒,且嵌合基因可以随病毒在细胞上稳定传代,但P1基因的替换在一定程度上影响了两嵌合病毒在BHK21细胞上的复制能力,使得嵌合病毒在BHK21上完全CPE时间长达19 h左右。2株嵌合病毒的成功拯救可为未来我国边境地区口蹄疫的有效防控提供坚实的技术储备。

摘  要:
To prevent the risk of trans-boundary transmission of new genotypic FMDV O/PanasiaⅡand A/Iran05 circulating in Asia regions and posing great menace to our national livestock husbandry.In this study,two chimeric viruses were constructed by replacing the whole P1 capsid protein coding region of O/HN/93 and A/GDMM/2013 infectious clones with the corresponding part of O/PanasiaⅡand A/Iran05,respectively.The recombinant viruses were proved to be rescued and competent for replication by indirect immunofluorescence,RT-PCR,gene sequencing,electronic microscopy,plaque phenotype and one-step growth curves.However,the recombinant viruses showed relatively lower ability of replication and longer time to reach complete CPE compared with parental viruses,being almost 19 hours.These recombinant FMDVs provide very useful materials for studying the vaccine strains against O/PanasiaⅡand A/Iran05 incursion.
