龙葵生物碱诱导HeLa细胞凋亡的研究

摘  要:
用MTT法观察龙葵生物碱的体外抗肿瘤作用,用TUNEL染色法检测龙葵生物碱对HeLa细胞凋亡的影响,用免疫细胞化学方法研究龙葵生物碱对HeLa细胞PCNA和突变型P53蛋白表达的影响。浓度为100μg/mL~800μg/mL的龙葵生物碱对HeLa细胞的生长均表现出一定的抑制作用;TUNEL染色结果提示400μg/mL的龙葵生物碱处理HeLa细胞48 h后,能够诱导更多的细胞出现凋亡;免疫细胞化学分析结果显示,400μg/mL的龙葵生物碱处理HeLa细胞48 h后,会使HeLa细胞中PCNA蛋白和突变型P53蛋白表达量显著下调。结果表明,龙葵生物碱在体外具显著抗宫颈癌活性,其作用机制是通过诱导更多的细胞出现凋亡而实现的。

摘  要:
MTT method was used to explore the anti-neoplastic action of solanine in vitro,TUNEL staining was used to analyze the apoptosis-inducing effect of solanine on HeLa cell line,and immumunocytochemistry method was further used to detect the effects of solanine on the protein expression of PCNA and mutant P53 in HeLa cell line.100 μg/mL-800 μg/mL solanine had certain inhibition effect on the growth of HeLa cell line.TUNEL staining results showed that after the treatment of solanine at a dose of 400 μg/mL for 48 h,more HeLa cells were induced to apoptosis.Immumunocytochemistry analysis showed that after the treatment of solanine at a dose of 400 μg/mL for 48 h,the expression of PCNA protein and the mutant P53 protein were all down-regulated in HeLa cells.These results indicated that solanine had significant anti-cervical cancer effect in vitro,and its anti-tumor mechanism be correlated with its cell apoptosis-inducing action.
