摘  要:
本研究选取NN DH系中的ZW46为母本,晚熟No.5246为父本(轮回亲本),利用cqDTFA7a位点候选区间及侧翼标记进行前景选择,其他18条染色体上的标记进行背景选择,通过连续回交及标记辅助选择方法构建甘蓝型春油菜早花位点cqDTFA7a的近等基因系.在BC2F1群体中,筛选到cqDTFA7a位点杂合的单株4株,背景回复率达到了95%以上,自交获得一个包含537单株的BC2F2群体,通过双亲基因组重测序,开发了8个InDel (insertion-deletion)标记,利用这些标记进一步加密了cqDTFA7a区间,将cqDTFA7a定位在IA7-4附近,相对遗传距离为0.1 c M,表型贡献率为47%,区间由原来的900 kb缩小到目前的300 kb.以上4个单株进一步回交获得前景位点杂合、背景回复率达到100%的单株4株,与No.5246构成了近等基因系,通过对上述4个单株自交获得一个包含843个单株的BC3F2群体,通过卡方检验,该群体花期遗传符合孟德尔遗传(3∶1),说明在该群体中,花期由一对显性基因控制.本研究为甘蓝型春油菜早花QTL位点cqDTFA7a的精细定位和候选基因挖掘奠定了基础.

摘  要:
In this study, ZW46 in the NN DH line was selected as the female parent, and late maturity No.5246 was used as the male parent(Recurrent parent). The cqDTFA7 a locus candidate interval and flanking markers were used for foreground selection, and the markers on the other 18 chromosomes were selected for background selection. The near-isogenic line of early flowering locus cqDTFA7 a in Brassica napus L. was constructed by continuous backcrossing and marker-assisted selection. In the BC2 F1 population, 4 plants with heterozygous cqDTFA7 a locus were screened, and the background recovery rate reached over 95%. Self-acquisition to obtain a BC2 F2 population containing 537 plants, and 8 InDels(insertion-deletion) mark were developed by re-sequencing of the parental genome. Use these markers to further encrypt the cqDTFA7 a interval, positioning cqDTFA7 a near IA7-4, the relative genetic distance is 0.1 c M, the phenotypic contribution rate is 47%, and the interval is reduced from the original 900 kb to the current 300 kb. The above four individuals were further backcrossed to obtain 4 plants per plant with heterozygous foreground sites and a background recovery rate of 100%, and building a near isogenic line with No.5246. Obtaining a BC3 F2 population containing 843 individuals by self-crossing the above four individuals,through the chi-square test, the flowering inheritance of this population is consistent with Mendelian inheritance(3∶1), indicating that in this population, the flowering stage was controlled by a pair of dominant genes. This study will laid the foundation for the fine mapping and candidate gene mining of early flowering QTL locus cqDTFA7 a in Brassica napus L..
