摘  要:
以建鲤(Cyprinus carpio var.jian)为研究对象,探讨丙氨酰-谷氨酰胺(Ala-Gln)对体外培养的免疫抑制建鲤淋巴细胞IL-lβ、IL-2以及HSP70mRNA表达的影响。通过对建鲤注射环磷酰胺(CY)来建立免疫抑制模型,以注射生理盐水的建鲤作为对照,分离外周血和头肾淋巴细胞在含不同Ala-Gln浓度(0.0、2.0、4.0、6.0、8.0和10.0 mmol/L)的培养液中进行体外培养。结果表明,Ala-Gln浓度为0.0、2.0、4.0mmol/L时,免疫抑制组建鲤外周血(PBL)IL-1β表达显著高于对照组(P<0.05),其他组差异不显著。对照组和免疫抑制组建鲤PBL IL-2表达均随Ala-Gln浓度增加而显著提高(P<0.05),且对照组均显著高于免疫抑制组(P<0.05)。免疫抑制组头肾淋巴细胞HSP70mRNA的表达随着Ala-Gln浓度的增加而显著增加(P<0.05),均显著高于对照组(P<0.05)。在本试验条件下,一定范围(4.0~10.0mmol/L)内添加Ala-Gln,可显著提高HSP70mRNA表达,降低血淋巴细胞IL-lβ表达的水平,恢复血淋巴细胞IL-2表达的水平,从而缓解免疫抑制。

摘  要:
To investigate the effects of Ala-Gln on IL-1β,IL-2 and HSP70 mRNA expression in Lymphocytes of Cyprinus carpio var.jian subjected to immunosuppression in vitro,the model of immunosuppression was established by injecting cyclophospamide,and the fish by injecting saline was taken as control.Lymphocytes of blood and head kidney were separated and cultured in media containingdifferent Ala-Gln concentrations(0.0,2.0,4.0,6.0,8.0and 10.0mmol/L).Results showed that the IL-1βexpression of immunosuppression group was significantly higher than that of control group at the range from 0.0~4.0mmol/L(P<0.05).There were no significant differences among the other Ala-Gln concentrations(P > 0.05).The IL-2and the HSP70 mRNA expression in the two group were both significantly increased with the increasing of Ala-Gln concentration(P<0.05),but the IL-2expression of control group was higher than that of the immunosuppression group(P<0.05)at every the Ala-Gln concentration.On the contrary,the HSP70 mRNA expression of immunosuppression group both higher than that of control group(P<0.05).In the present study,in a certain range of Ala-Gln(4.0 ~ 10.0mmol/L),Ala-Gln can promote the HSP70 mRNA expression,decrease the IL-1βexpression,upregulate the IL-2expression,then relieve the immunosuppression.
