猪霍乱沙门菌SC500运载TGEVS与猪IL-15融合基因疫苗的稳定性及免疫安全性

摘  要:
为研究猪霍乱沙门菌SC500作为TGEV S与IL-15融合基因疫苗pCC-S-IL-15、pCI-S-IL-15运载体的稳定性与免疫安全性,将外源表达质粒pCC-S-IL-15、pCI-S-IL-15及空载体质粒pCC-neo、pCI-neo转化SC500,采用体外增殖试验研究不同质粒在SC500中的稳定性,携带质粒的SC500对ST细胞的黏附率、侵袭力和增殖能力的影响;采用BALB/c小鼠口服免疫试验研究表达质粒在体内的稳定性及SC500对小鼠的安全性。结果显示:外源目的基因影响表达载体在运载体内的稳定性,就载体类型而言,对pCI-neo的影响大于对pCC-neo的影响;携带目的基因的外源质粒影响SC500对ST细胞的黏附率和侵袭率,pCC-neo影响大于pCI-neo;外源质粒影响SC500在小鼠体内的增殖能力,pCC-neo影响大于pCI-neo;SC500在体内的增殖中,其所运载的质粒具有一定的稳定性。结果表明,利用SC500运载基因疫苗pCC-S-IL-15、pCI-S-IL-15免疫小鼠具有相对的稳定性和免疫安全性。

摘  要:
To investigate the stability and safety of fusion-expression gene vaccines pCC-S-IL-15 and pCI-S-IL-15 with TGEV-S and porcine IL-15 genes using Salmonella choleraesuis SC500 strain as transporter,the exogenous expression plasmids pCC-S-IL-15,pCI-S-IL-15,empty plasmid pCC-neo and pCI-neo were transformed into SC500.The stability of plasmids in the transporter was studied by the in vitro proli-feration of SC500.The effects of SC500 carrying the plasmids on adhesiveness,invasiveness and proliferation of SC500 to ST cells was studied.The immunity experiments on BALB/c mice were carried out to investigate the stability in vivo of these plasmids and the security of SC500 in mice.The expression vector pCI-neo in the transporter was impacted stronger by exogenous genes than by pCC-neo.The ratio of adhesiveness and invasiveness of SC500 to ST cells was affected by exogenous plasmids and it was stronger for pCC-neo than pCI-neo.Exogenous plasmids influenced the reproductive activity of SC500 in vivo.As to the type of the vectors,the action of pCC-neo was more powerful than pCI-neo.The plasmids transported into SC500 possessed certain stability when SC500 was proliferated in vivo.The results showed that SC500 was safer and steadier than the transporter of pCC-S-IL-15 and pCI-S-IL-15.
