猬迭宫绦虫广东分离株线粒体pcox1基因的克隆及序列分析

摘  要:
为阐明猬迭宫绦虫广东分离株的遗传变异情况,本研究利用PCR技术扩增了猬迭宫绦虫的部分细胞色素C氧化酶第Ⅰ亚基(pcox1)基因,经SSCP筛选出带型差异的代表样品进行测序,经系统发育树表明,广东分离株与已知猬迭宫绦虫位于同一分枝。同时研究表明猬迭宫绦虫pcox1序列种内相对保守,又存在一定种间差异,可作为种间遗传变异研究的标记,研究结果为进一步研究猬迭宫绦虫的群体遗传结构奠定了基础。

摘  要:
To examine sequence variation in the mitochondrial cytochrome c oxidase subunit 1 (cox1) gene among Spirometra erinaceieuropaei isolates from Guangdong area. Representative samples were selected by analysis of single-strand conformation polymorphism (SSCP) and were subjected to sequencing and comparison with relevant sequences available in GenBank. The partial cox1 (pcox1) sequences amplified from all S. erinaceieuropaei had same length of 396 bp and shared more than 98 % sequence similarity between Guangdong isolates and the S . erinaceieuropaei in GenBank , while the similarity between S. erinaceieuropaei and the other cestode was less than 85 %. The phylogenetic tree showed that the Guangdong isolates and other S. erinaceieuropaei were clustered in the same clade. These findings showed that inter-specific sequence differences were significantly greater than intra-specific sequence differences in pcox1, indicating that pcox1 sequence could be used as genetic marker for the specific differentiation among cestodes and for studying genetic variation within S. erinaceieuropaei.
