波纹唇鱼雄激素受体基因的克隆与表达分析

摘  要:
通过RT-PCR克隆并得到了波纹唇鱼雄激素受体基因的全长,共3514bp,编码755个氨基酸。氨基酸序列分析表明,该序列有3个结构域,即转录激活区、DNA结合区和配体结合区。系统发育分析表明,波纹唇鱼雄激素受体与贝氏隆头鱼、西氏拟隆头鱼和欧洲舌齿鲈的雄激素受体具有较高的相似性,分别为85.0%、82.5%和79.5%。组织表达分析表明,波纹唇鱼雄激素受体基因在波纹唇鱼的前脑、中脑、后脑、垂体、下丘脑、心脏、肝脏、性腺、脾脏、肾脏、前肠、后肠、肌肉中均有表达,表达量有所差异,肾脏中表达量最为丰富,前肠中表达量最少。波纹唇鱼雄激素受体基因的成功克隆与分析,为波纹唇鱼生殖调控的研究提供了理论基础。

摘  要:
The full length cDNA sequence of androgen receptor(ar)in humpheaded wrasse Cheilinus undulatus was cloned by RT-PCR and RACE.The cDNA sequence of ar was found to be 3514 bp,encoding755 amino acids(aa),and amino acid sequence analysis showed that the deduced amino acid sequence of ar contained all three domains characteristics of ars,transcriptional activation domain,DNA-binding domain and ligand-binding domain.Phylogenetic tree indicated that the ar of humpheaded wrasse had high homology of 85.0% with Labrusbergylta,of 82.5% with Pseudolabrussieboldiand of 79.5% with Dicentrarchus labrax.The ar was expressed in brain area,heart,liver,gonad,spleen,kidney,foregut,hindgut and muscle,with the maximal expression in the kidney,and the minimal in the foregut.The successful cloning and analysis of ar from humpheaded wrasse provide a theoretical for the study of reproductive regulation.
