rhG-CSF对大肠杆菌性乳房炎昆明小鼠血液生理指标和乳腺组织形态的影响

摘  要:
将120只泌乳8~10d的初产昆明小鼠,随机分为5组。Ⅰ,Ⅱ组皮下注射0.1mL生理盐水,Ⅲ,Ⅳ,Ⅴ组分别按10,20,30μg/kg体质量注射0.1mL rhG-CSF;24h后,Ⅰ组第4对乳房每侧灌注25μL生理盐水,Ⅱ~Ⅴ组灌注等量含1.5×107 CFU/mL大肠杆菌菌液。于感染后0,24,72,120h,每组选取6只小鼠,检测血液白细胞数量、血清TNF-α含量、乳腺组织大肠杆菌数量并观察组织病理学变化。结果显示,整个试验期间,Ⅴ组血液中白细胞总数及其分类均显著高于Ⅰ,Ⅱ,Ⅲ,Ⅳ组,Ⅳ组高于Ⅱ,Ⅲ组,Ⅲ组高于Ⅰ,Ⅱ组,Ⅱ组高于Ⅰ组;血清中TNF-α含量比较,24h时,Ⅱ组显著高于Ⅰ,Ⅲ,Ⅳ,Ⅴ组,72和120h时,Ⅱ,Ⅲ,Ⅳ,Ⅴ组显著高于Ⅰ组,Ⅱ,Ⅲ组显著高于Ⅴ组;120h时,Ⅴ组未检出大肠杆菌,且乳腺组织无明显炎症损伤,Ⅱ组大肠杆菌数量极显著高于Ⅲ,Ⅳ组,Ⅲ组极显著高于Ⅳ组,其中Ⅱ组小鼠乳腺腔和间质内大量炎性细胞浸润,组织结构紊乱,Ⅲ组有少量炎性细胞浸润,Ⅳ组组织结构完整,组织损伤轻微。结果表明,rhG-CSF能通过升高小鼠血液中白细胞数量和TNF-α浓度,达到提高病原菌清除率,从而减轻组织损伤的目的,且以30μg/kg效果最佳。

摘  要:
To explore recombinant granulocyte colony-stimulating factor(rhG-CSF)on the prevention effect of mastitis,mammary 8-10 days 120 mice were randomly divided into five groups.Ⅰ,Ⅱgroup of subcutaneous injection of 0.1 mL saline;groupⅢ,ⅣandⅤ,respectively injection by 10,20 and 30μg/kg bodyweight 0.1 mL rhG-CSF.After injecting 24 h,theⅠ group was given 50μL normal saline,and the Ⅱ,Ⅲ,Ⅳ and Ⅴ were given 50μL E.coli at concentration of 1.5×107 CFU/mL.After infection in 0,24,72,120 h,sampling of randomly selected 6 mice in each group,to detect the contents of blood serum TNF-α,the number of E.coli and histopathological changes of mammary gland.The results showed that total number of white blood cells in the blood and classification decreased,theⅤ group was significantly higher than that of groupⅠ,Ⅱ,Ⅲ,Ⅳ;comparison of serum TNF-αcontent at 24 h,Ⅱ group was significantly higher than theⅠ,Ⅲ,Ⅳ and Ⅴgroup,at 72 and 120 h,Ⅱ,Ⅲ,Ⅳ andⅤ group was significantly higher than group Ⅰ;at 20 h,Ⅴgroup without detection of E.coli,and no obvious inflammation of the breast tissue in groupⅡthe number of E.coli was significantly higher than that of group Ⅲ and Ⅳ,Ⅱ group mice mammary gland cavity and stroma were observed a large number of inflammatory cells infiltration,disorder of organizational structure,groupⅢ had a small amount of inflammatory cells infiltration,Ⅳ group structure full,injury minor.The rhG-CSF by increasing the number of white blood cells in the blood of mice improves the clearance rate of bacteria,so as to reduce tissue damage,and 30μg/kg to best effect.
