中间锦鸡儿WRKY15基因克隆及其生物信息学分析

摘  要:
WRKY家族是高等植物中最大的转录因子家族之一,参与多种生物与非生物胁迫过程的调节.本研究采用PCR技术从中间锦鸡儿(Caragana intermedia)中克隆了CiWRKY15基因.结合生物信息学方法对Ci-WRKY15基因编码的蛋白质进行理化性质、亚细胞定位、蛋白结构以及系统进化等进行预测分析.结果显示:Ci-WRKY15基因序列包含1005bp开放阅读框,编码335个氨基酸,含有一个典型的WRKY结构域,属于Group IId亚族.预测蛋白分子量为47.39KD,等电点为8.59,它的亚细胞定位于细胞核内.该研究结果为进一步挖掘与非生物胁迫相关的WRKY15转录因子的功能,以及为改进植物抗生物和非生物逆境的特性提供了理论依据.

摘  要:
In this study, WRKY15 gene was cloned from the cDNA of Caragana intermedia by PCR method. Bioinformatics analysis was used to predict the physico-chemical properties, subcellular localization, protein structure and phylogenetic evolution of the protein encoded by CiWRKY15 gene. The results showed that the CiWRKY15 gene sequence contained 1005 bp open reading frame, encoded 335 amino acids, and contained a typical WRKY domain, belonging to Group IId subgroup. The predicted molecular weight of putative protein was 47.39 KD and its isoelectric point was 8.59. Its subcellular localization was in the nucleus. The results of this study provide a theoretical basis for further exploring the function of WRKY15 transcription factors related to abiotic stress and improving the characteristics of plant resistance to biotic and abiotic stress.
