大孔吸附树脂分离纯化千斤拔总黄酮的工艺研究

摘  要:
为研究大孔树脂分离纯化千斤拔总黄酮的最佳工艺,以总黄酮的含量为指标,通过静态吸附解析试验比较7种不同类型大孔吸附树脂的吸附解析特性,确定AB-8型大孔吸附树脂适用于千斤拔总黄酮的分离纯化。通过动态吸附试验确定了大孔吸附树脂分离纯化千斤拔总黄酮的最佳工艺条件。结果表明:大孔树脂分离纯化千斤拔总黄酮的最佳工艺为:上样液质量浓度相当于原生药质量浓度为0.12g·mL-1,最大上样量为12.83mg·mL-1,上样液的pH为5.0,上样流速为2.0mL·min-1,洗脱液乙醇体积分数为70%,洗脱剂用量为7BV,洗脱流速为1.5mL·min-1。在此条件下,千斤拔总黄酮的纯度由31.26%提高至65.7%,说明该工艺稳定可靠,可用来分离纯化千斤拔总黄酮。

摘  要:
To optimize the process of separating and purifying total flavones from Moghania philippinensis by macroporous adsorption resins.The adsorption and desorption of characteristics of seven different types of macroporous was investigated with content of total flavones as the evaluation index.AB-8resins showed the best effect of separation and purification of total flavones from Moghania philippinensis.Optimum conditions of separation and purification of total flavonoids by the macroporous resin was determined according the dynamic adsorption experiments.The optimum process conditions was as follows:the loaded sample concentration was 0.12g·mL-1,the maximum sample volume was 12.83 mg·mL-1,pH was 5.0,loaded velocity was2.0mL·min-1,elution solvent concentration was 70%ethanol,elution solvent volumew was 7BV and desorption velocity was 1.5mL·min-1.Under the process conditions,the purity of total flavones from Moghania philippinensis raised to 65.7%from 31.26%.The process could be used to separat and purif the total flavonoids from Moghania philippinensis.
