日粮不同能量水平对乌金猪脂肪组织Leptin和RBP4基因表达的影响

摘  要:
研究日粮不同能量水平对乌金猪脂肪组织中脂肪细胞分泌因子leptin和RBP4基因表达的影响。选取体重约15 kg的乌金猪54头,随机分为3组,分别饲喂消化能为14.22,12.89,11.74 MJ/kg的日粮,在30,60,100 kg体重时屠宰,分别测定血液中胰岛素和葡萄糖含量及胴体脂肪沉积率,取皮下脂肪组织用荧光定量PCR检测leptin和RBP4基因的表达水平。结果显示,饲喂高能量日粮乌金猪胴体脂肪沉积率和血液胰岛素含量高于饲喂低能量日粮组(P<0.05),日粮能量水平对血液葡萄糖含量无明显影响(P>0.05);脂肪组织中leptin基因的表达水平升高(P<0.05),RBP4基因的表达水平降低(P<0.05)。表明高能量日粮通过上调leptin基因的表达、下调RBP4基因的表达,和胰岛素的作用促进脂肪沉积。

摘  要:
The experiments were performed to identify the effect of dietary different energy levels on leptin and RBP4 gene expression in adipose tissue of Wujin pigs.Fifty-four Wujin pigs were randomly assigned into 3 groups and feed on dietary contains three different energy levels,which were low energy(DE: 11.74 MJ/kg),middle energy(DE: 12.89 MJ/kg) and high energy(DE: 14.22 MJ/kg) respectively.At the body weights of 30,60 and 100 kg,Wujin pigs were slaughtered and subcutaneous adipose tissue were taken for analyzing leptin and RBP4 gene expression by real-time PCR,the contents of insulin and glucose in blood and carcass fat deposition percentage were detected,respectively.The results showed that feed higher dietary energy could increase carcass fat deposition percentage and blood insulin content(P<0.05),increase leptin gene expression(P<0.05),decrease RBP4 gene expression(P<0.05),but had no influence on blood glucose content(P>0.05).This suggested that higher dietary energy could promote carcass fat deposition by up-regulating leptin gene expression,down-regulating RBP4 gene expression,and through the action of insulin.
