载入骨形态发生蛋白的羟基磷灰石/丝素蛋白复合支架的细胞相容性研究

摘  要:
骨形态发生蛋白-2(BMP-2)能够促进成骨细胞分化和骨组织的再生。将制备的羟基磷灰石/丝素蛋白(HA/SF)复合支架在50μg/mL BMP-2溶液中浸渍4 h后,获得了载入BMP-2的HA/SF复合支架。体外检测在原核表达系统中表达、纯化的BMP-2有促进细胞增殖的生物活性;BMP-2从HA/SF复合支架中的释放可持续9 d,并保持其生物活性,其中BMP-2在第1天的释放率即达到了43%;细胞实验结果显示,与空白HA/SF复合支架相比,载入BMP-2的HA/SF复合支架可以促进人骨肉瘤细胞(MG-63)的增殖与分化;RT-PCR检测BMP-2的加入使细胞内骨钙素基因mRNA转录水平明显提高,即骨钙素基因的表达增强。研究结果提示:HA/SF复合支架可吸附BMP-2并使其缓慢释放,从而促进细胞的增殖与分化,具有良好的细胞相容性,是一种较理想的骨组织缺损修复材料。

摘  要:
Bone morphogenetic protein-2(BMP-2) plays an important role in stimulating the osteoblast differentiation and bone formation.In this study,the hydroxyapatite/silk fibrion(HA/SF) composite scaffolds were fabricated,which were then soaked in a 50 μg/mL BMP-2 solution for 4 h to obtain HA/SF composite scaffolds carrying BMP-2 protein.The analysis of biological activity of the BMP-2,which was expressed in Escherichia coli expression system and purified in vitro,showed that BMP-2 could promote cell proliferation.Release of BMP-2 from HA/SF composite scaffold could last 9 d and still maintain its biological activity,during which the release rate of BMP-2 reached 43% in the first day.Results of cell culture experiment showed that,compared to the blank HA/SF composite scaffold,the HA/SF composite scaffold carrying BMP-2 could promote proliferation and differentiation of human osteosarcoma cells(MG-63).RT-PCR detection showed that the mRNA transcriptional level of cellular osteocalcin gene was increased obviously by addition of BMP-2,that is,expression of osteocalcin gene was enhanced.These results suggested that the HA/SF composite scaffold can adsorb BMP-2 and slowly release it,which promotes cell proliferation and differentiation and has good cell biocompatibility,being a good material for bone tissue repair.
