A蛋白-金颗粒复合物(pAg)诊断和检测植物病毒的研究

摘  要:
以A蛋白-金颗粒复合物(Protein A—Gold complexes)标记植物病毒粒体。利用抗原抗体特异性反应和A蛋白与抗体中免疫球蛋白IgG结合的特性,在电镜下观察到同源关系中的金颗粒吸附于病毒粒体周围;而异源关系中,金颗粒则被冲洗掉或随机分布。该法专化性受抗血清稀释度和吸附时间等因素影响。对杆状、线状病毒粒体来说,抗血清最适稀释度为1×10~2—10~3倍,对球形病毒粒体来说,抗血清稀释度以1.2×10~3倍为佳。抗血清吸附时间以3—10分钟的效果最好。

摘  要:
Several plant viruses were labelled with protein A-gold complexes. Under electron microscopy it was observed that gold particles were absorbed around virus when homologous antibody was used, whereas, gold particles were randomly distributed or couldn't be linked when heterologous antibody was used. The specificity of the method was affected by several factors, such as dilution of antibody and antibody incubation duration. The bust results were obtained when antibody was diluted 100-1000 times for elongated viruses and 1200 for spherical viruses. Optimum antibody incubation duration was 3-10 minutes.
