免疫球蛋白及其受体在感染禽致病性大肠杆菌鸡法氏囊中的表达水平分析

摘  要:
为研究鸡人工感染禽致病性大肠杆菌后免疫球蛋白及其受体在感染鸡群和非感染正常鸡群中的表达差异,将32只4周龄AA商品肉鸡随机分成两组,16只作为感染组,16只作为对照组。感染组左胸气囊注射108PFU的禽大肠杆菌O1∶K1∶H7 0.1 mL/只;对照组注射同等剂量的磷酸盐缓冲盐水,两组鸡严格隔离,在相同条件下的负压隔离器中饲养。攻毒后1 d,两种各选取8只进行剖杀,攻毒后5 d,剖杀剩余所有试验鸡,采集法氏囊样本检测免疫球蛋白及其受体基因表达水平。结果表明:在攻毒后5 d,鸡法氏囊中免疫球蛋白受体的表达水平显著下调,而免疫球蛋白A、M、Y均未发生显著性差异表达。在攻毒后1 d,所有候选基因均未发现显著性差异表达。结果提示:在禽大肠杆菌感染过程中免疫球蛋白受体表达量的增加可能比免疫球蛋白基因表达总数的增多更重要。研究从分子水平上分析了免疫球蛋白及其受体在禽大肠杆菌感染过程中的免疫应答反应,为深入了解宿主抵御禽大肠杆菌感染的作用机制奠定了重要基础。

摘  要:
To study the expression of immunoglobulins(Ig)and receptors of birds challenged with avian pathogenic Escherichia coli(APEC)and non-challenged birds,thirty two four-week-age commercial broilers were randomly divided into two groups,sixteen birds in challenged group and sixteen birds in control group. Challenged group was injected 108plaque-forming units(PFU)of APEC O1∶K1∶H7 with 0.1 mL per bird,non-challenged control group was injected with the same dosage of phosphate buffered saline(0.1 m L per bird). The two groups were housed in separate cages in the same filtered-air,positive-pressure isolation rooms. After 1 day post-infection(dpi),eight birds were necropsy in each group. After 5 dpi,all birds were necropsy. Bursa were collected to identify the expression level of Igs and the receptor.The results showed that the expression of Ig receptor was significantly down-regulated in infected birds while Ig A,Ig M,and Ig Y were not detected differential expression in control group after 5 dpi. All the detected genes were not significantly differentially expressed in each group after 1 dpi. This results indicated that increased expression of Ig receptor in the bursa might be more important than up-regulating the total number of Ig class genes because the receptor did not require any additional specificity. This study measured the immune response of Igs and receptor at molecular level in host during APEC infection providing an important foundation in understanding the mechanism of host immune response to APEC infection.
