樟叶越桔愈伤组织诱导及其细胞悬浮培养体系建立

摘  要:
以樟叶越桔组培苗幼嫩叶片为外植体,研究了植物生长调节剂对其愈伤组织诱导和增殖的影响,建立樟叶越桔的细胞悬浮培养体系。结果表明,樟叶越桔的愈伤组织诱导的适宜培养基为WPM+2.0mg/L 6-BA+0.05mg/L NAA+2.0 mg/L 2,4-D+30%蔗糖。樟叶越桔愈伤组织增殖的最佳植物生长调节剂组合为2.0mg/L ZT+0.4mg/L NAA,得到的蓬松愈伤组织适宜进行其悬浮培养。WPM 3/5有机为适宜的改良培养基,通过细胞悬浮生长曲线的测定,确定樟叶越桔细胞悬浮培养较适宜的继代周期为14d,最长不宜超过21d。

摘  要:
Using the young leaves of the Vaccinium dunalianum seedlings as an explant,the effects of plant growth regulators on the induction and proliferation of callus were investigated for the establishment of the cell suspension culture system.The results showed that the optimal medium for callus induction were WPM+2.0 mg/L 6-BA+0.05 mg/L NAA+2.0 mg/L 2,4-D+30% sucrose.The best plant growth regulator combination for callus proliferation was 2.0 mg/L ZT+0.4 mg/L NAA,and the obtained callus was suitable for cell suspension culture.The best improved medium was organic WPM 3/5,and the callus growth curve test showed that the suitable subculture period was 14 d and the longest subculture period was not more than 21 d.
