番茄抗叶霉病基因Cf-12定位及候选基因分析

摘  要:
近年来,番茄叶霉病的大面积发生对中国番茄生产造成极大影响。叶霉病菌(Cladosporium fulvum)与番茄植株发生的亲和性互作是番茄叶霉病发生的根源,以叶霉病抗性基因Cf为主导的抗叶霉病育种研究是工作的重点,但由于Cf基因抗性不断被克服,急需挖掘抗性更广更强的Cf基因。番茄品种‘CGN7495’含有Cf-12基因,田间表现出有效的C. fulvum抗性。本研究通过构建6世代群体,分析了Cf-12抗性的遗传规律,证实了该基因符合单基因的显性遗传规律,为分离群体分组分析法筛选Cf-12基因做了铺垫;采用父母本重测序及F2代极端性状混池简化测序SLAF (Specific locus amplified fragment sequencing)来定位Cf-12基因,将其定位到番茄第6号染色体上并最终筛选出3个候选基因。研究为Cf-12的精确克隆和相关抗病机制的研究提供了理论参考。

摘  要:
In recent years, the large area of the tomato leaf mold has a great effect on the production of tomato in China. The compatible interaction between C. fulvum and tomato plants is the main reason for the occurrence of tomato leaf mold. Therefore, the resistance Cf breeding work is the key point of our work to control leaf mold. As the resistance of Cf genes is overcomed continuously, it is urgent to mine new resistance Cf genes. The material'CGN7495' contains Cf-12 gene and shows effective resistance to C. fulvum in the field. By the six generations, The article confirmed that Cf-12 accorded with the dominant inheritance of single gene. Pave the way for screening Cf-12 gene by using the separation group analysis method. This study used parental resequencing methods and extreme trait pool of F2 SLAF(Specific locus amplified fragment sequencing) to locate the Cf-12 gene and the gene was mapped to chromosome 6 of tomato. As a result, three candidate genes were successfully screened. The study provided a theoretical reference for the study of the precise cloning of Cf-12 and the related disease-resistant mechanism.
