摘 要:
以羽衣甘蓝(Brassica oleracea var.acephala)两个自交系(F6代)作比较,利用ISSR标记技术检测了羽衣甘蓝3个DH1代株系的遗传稳定性,并对5个群体进行聚类分析。结果表明,3个DH1代株系DN31、DP70、DT2和两个自交系B62、B111利用9条ISSR引物分别扩增出55、51、50、54和51条清晰条带,其中,多态性位点DN31有3个,DP70有2个,DT2没有,B62有19个,B111有15个。羽衣甘蓝DH1代株系内基本不存在遗传差异,基因型是纯合的,两个自交系遗传稳定性相对较差。表明小孢子培养技术在获得纯合体材料上比人工自交的方法高效、快捷,而且获得的纯系基因型更纯、更稳定。

摘 要:
To compare two inbred lines(F6) of ornamental kale(Brassica oleracea var.acephala),genetic stability of each individual plant in three DH1 lines derived from the two inbred lines was detected by ISSR marker,and clustering analysis was performed in the five populations by use of system clustering method.The results showed that 3,2,0,19,15 polymorphic loci had been detected out respectively from 55,51,50,54,51 loci in 3 DH1 lines(DN31,DP70,DT2) and 2 inbred lines(B62,B111) using 9 ISSR primers.Almost no genetic difference had been detected in 3 DH1 lines,but several genetic differences had been detected in 2 inbred lines.This indicated that genotypes of DH1 lines derived from microspore culture were homozygous.It showed that using the microspore culture technique to obtain homozygote materials was more efficient and faster than using artificial hybridization,and genotypes obtained by microspore culture technique were more pure and stable.
