小球藻Chlorella sp. MEM25的分离鉴定及耐受性试验

摘  要:
从盐卤水样品中分离出一株藻种MEM25,经形态学检验和分子生物学鉴定为小球藻Chlorella sp..以IMET1作为对照组对MEM25进行耐受性试验,结果表明,在不同盐度、光照、温度条件下MEM25的D750 nm和Fv/Fm均高于IMET1.当试验条件分别为盐度7.0%、光照250μmol·m-2·s-1、温度35℃条件时,MEM25的D750 nm分别为初始浓度的12.2、5.2、10.6倍.在模拟户外条件培养试验中MEM25的干重为1.88 g·L-1,蛋白质含量为53%,分别是IMET1的2.58和1.66倍.在户外培养试验中MEM25的最高浓度为12.65×107个·mL-1,最大日增长量为3.31×107个·mL-1,细胞中8种必需氨基酸含量占总氨基酸含量的49.5%,必需脂肪酸C18∶3占总脂含量的34.7%.

摘  要:
A marine microalgae MEM25 was isolated from brine samples and was identified as Chlorella sp. via morphological analysis and molecular biology techniques. And IMET1 was used as the control group to evaluate the tolerance of MEM25. The results showed that the D_(750 nm) and Fv/Fm values of MEM25 were higher than those of IMET1 under different salinities, illuminations and temperatures. Under 7.0 % salinity, 250 μmol·m~(-2)·s~(-1) illumination and 35 ℃, the D_(750 nm) values of MEM25 were 12.2, 5.2 and 10.6 times higher than those of the initial values, respectively. The dry weight and protein content of MEM25 reached 1.88 g·L~(-1) and 53% during simulated outdoor cultivation, which were 2.58 and 1.66 times more than those of IMET1, respectively. The maximum concentration of MEM25 cells was 12.65×10~7 individual·mL~(-1) and the maximum daily growth was 3.31×10~7 individual·mL~(-1) in outdoor culture. Total essential amino acids in MEM25 accounted for 49.5% of the total amino acids and the essential fatty acid C18∶3 accounted for 34.7% of the total lipids.
