四种野生梨离体培养体系的建立及耐盐性比较

摘  要:
以杜梨、豆梨、川梨和山梨为试材,进行了无菌体系的建立和增殖培养基的优化,在此基础上进行了Na盐胁迫下耐盐相关生理指标的测定,以期进行不同材料耐盐性的比较,探讨利用离体培养体系进行耐盐性鉴定的可行性。结果表明:不同种类梨芽的增殖对培养基中附加的各类生长调节剂的配比要求不同,筛选得到最佳芽增殖培养基为杜梨MS+6-BA 2.0mg/L+IBA 0.1mg/L+GA30.5mg/L;豆梨MS+6-BA 2.0mg/L+IBA 0.3mg/L+GA30.1mg/L;川梨MS+6-BA 2.0 mg/L+IBA 0.1 mg/L+GA3 0.5 mg/L;山梨MS+6-BA 1.0 mg/L+IBA0.3mg/L+GA30.5mg/L。对4种梨试管苗进行不同浓度NaCl胁迫处理的结果表明,不同梨盐害指数从高到低分别为川梨、豆梨、山梨、杜梨;对保护性酶类的活性及脯氨酸含量测定分析显示,4种梨中超氧化物歧化酶(SOD)、过氧化物酶(POD)、过氧化氢酶(CAT)和脯氨酸(Pro)在NaCl处理浓度较低时均呈现上升趋势,杜梨的活性变化相对平缓,而MDA变化以川梨的活性最高。

摘  要:
Using orthogonal design methods,the suitable medium used for bud proliferation of Pyrus betulaefolia,P.calleryana,P.pashiaand P.ussuriensis were screened and the salt tolerance were analysed and compared.The results showed that the suitable medium for the bud proliferation and growth of P.betulaefolia,P.calleryana,P.pashiaand P.ussuriensis were MS+6-BA 2.0mg/L+IBA 0.1mg/L+GA30.5mg/L,MS+6-BA 2.0mg/L+IBA 0.3mg/L+GA30.1mg/L,MS+6-BA 2.0mg/L+IBA 0.1mg/L+GA30.5mg/L,MS+6-BA 1.0mg/L+IBA 0.3mg/L+GA30.5mg/L,respectively.The salt injury index of plantlets treated with different concentration of NaCl and observed after four weeks revealed that the result as the following P.pashia>P.calleryana>P.ussuriensis>P.betulaefolia.The activity of SOD,POD,CAT and the content of Pro showed all rising in 4species of wild pear when the medium added lower concentration of NaCl,and there was a slowly change trend in P.betulaefolia.In the change of the content of MDA,the concentration of P.pashia was the highest.
