MIκBα对TNF-α诱导的大鼠肝星状细胞凋亡的影响

摘 要:
目的:探讨MIκBα对TNF-α诱导的大鼠肝星状细胞生长抑制和凋亡的影响。方法:首先用不同剂量的TNF-α对培养成功的MIκBα稳定转染组细胞和未转染大鼠肝星状细胞进行诱导,并使用间接免疫荧光法检测细胞中NF-κB/P65蛋白的表达,应用MTT法测定细胞生长率,然后用Annexin V-FITC和PI试剂分别进行双标记利用流式细胞仪对两组细胞的凋亡情况进行检测。结果:转染MIκBα的大鼠肝星状细胞的组NF-κB蛋白的表达明显被抑制,转染组较未转染组的细胞生长明显受到抑制,两者比较有统计学差异性。转染MIκBα的大鼠肝星状细胞有较高的凋亡率,而未转染MIκBα的大鼠肝星状细胞只有少许细胞发生凋亡,两者比较差异有统计学意义。结论:MIκBα表达可以抑制NF-κB蛋白的表达,从而提高大鼠肝星状细胞对TNF-α的敏感性,通过诱导肝星状细胞的凋亡,可能寻找到一条治疗肝纤维化的途径。

摘 要:
Objective:To explore the role of expression of MIκBα in TNF-αinduced apoptosis in rats hepatic stellate cell in vitro.Methods:A mutant IκBα was transfected into HSC-T6 cells by lipofectin transfection technique and its transient effect was examined 48h after transfection.The activation of NFκB was detected by immune fluorescence cytochemistry and western blot methods with anti-p65 antibody.The apoptosis inhibiting rate induced by TNF-α in both transfected and untransfected rat hepatic stellate cells were measured respectively by MTT methods.Labeled with Annexin V-FITC and PI kits,the apoptotic cells in two groups were detected by the flow cytometry.Results TNF-α could activate NFκB IN untransfected cells but not transfected rat hepatic stellate cells.The percentages of apoptosis in transfected cells were significantly higher than in untransfected ones(P<0.01).And so did the inhabiting rate(P<0.01).Conclusion The resistance of rat hepatic stellate cells towards apoptosis induced by TNF-α can be mediated by NF-κB activation.The inhibition of NF-κB activation by mutant IκBα can attenuate the resistance of rat hepatic stellate cells and increase its sensitivity to TNF-α.The way to treat liver fibrosis can be sought through apoptosis of rat hepatic stellate cells.
