史密斯桉愈伤组织的诱导及分化

摘  要:
通过研究史密斯桉愈伤组织诱导及分化成苗,发现以史密斯桉老茎断面萌发的新枝茎尖第1~7节作外植体,使用0.2%HgCl2消毒2~3min效果较好;2,4-D对史密斯桉愈伤组织的形成是必要的,使用1/2MS+2,4-D1.5mg·L-1+KT1.5mg·L-1或1/2MS+2,4-D1.5mg·L-1+ZET1.5mg·L-1能较好诱导愈伤组织形成;使用1/2MS+6-BA2.0mg·L-1+NAA0.2mg·L-1培养基能较好诱导茎形成的愈伤组织分化成芽,进而成苗,诱导率虽低(仅为20%),但增殖系数较高(高达5~10);叶形成的愈伤组织不能分化成芽;NAA与细胞分裂素配比诱导愈伤组织分化成芽优于IAA、IBA;6-BA诱导史密斯桉愈伤组织成苗效果最好,其最佳质量浓度为1.0~2.0mg·L-1;0.5~2.0mg·L-1的GA对史密斯桉苗的伸长有显著作用;20mg·L-1的Vc或0.5%活性炭能明显减轻史密斯桉愈伤组织的褐化作用。

摘  要:
The callus of Eucalyptus smithii L. was induced to form using the 1st to 7th nodes of shoot tip germinated from old stems as explants, 0.2%HgCl_2 disinfecting for two to three minutes, 1/2MS+2,4-D1.5mg·L~ -1 +KT1.5mg·L~ -1 or 1/2MS+2,4-D1.5mg·L~ -1 +ZET1.5mg·L~ -1 as culture media. The callus from stem was induced to form adventitious buds when using 1/2MS+6-BA2.0mg·L~ -1 +NAA0.2mg·L~ -1 as culture medium. Although the inducement rate is low, the coefficient of reproduction is high. The callus from leaf could not induce to form adventitious buds. 1.0~2.0g/L is the optimal concentration for 6-BA to induce callus to form adventitious buds, and 0.5~2.0g/L of GA has an obvious effect on seedling elongation. Vc at a concentration of 20mg/L or 0.5% of activated carbon can lighten browning effectively.
