苯酚-次氯酸钠比色法测定土壤脲酶活性影响因素的研究

摘  要:
土壤脲酶常采用苯酚-次氯酸钠比色法测定,该法测定结果重现性好,应用广泛,但存在培养土壤后滤液浑浊、带色的现象,影响测定结果准确性。参照此法,用KCl+HCl代替甲苯,选取3种代表性的土壤作为研究对象,针对储存方式、培养条件、提取剂浓度等重要参数进行考察,以期获得较高的准确性。实验结果表明,培养12 h、培养温度25℃,1.0 mol/L KCl+0.01mol/L HCl的浸提剂提取,其结果是传统方法的1.2~1.5倍;新鲜土壤-20℃或4℃贮存,测定结果无明显差异;风干土较新鲜土脲酶活性降低15%~30%。本文是对现有研究方法的补充,对脲酶活性测定结果的准确性、研究数据的可比性,有现实的参考价值。

摘  要:
Phenol-sodium hypochlorite colorimetric method with a better reproducibility, is the most widely used to measure the urease activities. However, the filtrate of cultured soil is turbid and colored, which affects the accuracy of the determiner results. In order to improve the accuracy of this method, toluene was replaced with KCl + HCl and the measured results for the urease activities of three representative soils under four crucial parameters, including storage mode, condition of cultivation, and concentration of extractant. The results showed that compared with the traditional method, the measured results were 1.2-1.5 times better under the conditions of 12 h extraction, 25 ℃ culture temperature, and 1.0 mol L-1 KCl and 0.01 mol L-1 HCl extract. There was no significant difference in the determination results between the fresh soil storage at-20 ℃ and 4 ℃. The urease activities of the air-dried soil were reduced by 15% to 30% compared with the fresh soil. The research results could be a supplement for the existing method and provide realistic references for the determination accuracy of urease activities and the comparability of the research data.
