带叶兜兰壮苗生根培养基及移栽基质选择

摘  要:
以带叶兜兰(Paphiopedilum hirsutissimum)组培苗为外植体,研究5种培养基对带叶兜兰组培苗壮苗生根的影响,设计6种基质处理,以筛选组培苗出瓶后的最佳移栽基质。结果表明:初步筛选出较适宜壮苗生根的培养基为1/2MS+NAH2PO4170 mg/L+NAA 0.1 mg/L,植株生物量增加显著;随着NAA浓度的增加,植株的壮苗生根效果受到不同程度的抑制。单独利用腐熟过的碎树皮做基质,试管苗移栽成活率均达99.7%以上。

摘  要:
Taking tissue culture seedling of Paphiopedilum hirsutissimum as tested material, the effects of fivedifferent mediums on P. hirsutissimum seedling growth and rooting were investigated, and six differenttransplanting matrix treatments were designed to select the best one for the transplanting of tissue cultureseedling. The results showed that 1/2MS+NAH2PO4 170 mg/L+NAA 0.1 mg/L was the optimum medium for theseedling growth and rooting, and the plant biomass increased significantly. With the increase of NAAconcentration,plant seedling rooting was inhibited. The maturity of the crushed bark was the optimized matrixfor the in vitro plantlet of P. hirsutissimum transplantation, which produced higher survival rate(>99.7%).
