带叶兜兰非共生萌发试验

摘  要:
以带叶兜兰同种异株授粉的果实为试验材料,采用正交实验设计,研究了非共生条件对带叶兜兰种子萌发的影响,以期为带叶兜兰的快速繁殖并最终实现产业化生产提供技术参考。结果表明:基本培养基的种类是带叶兜兰非共生萌发的主要影响因素,初步筛选出种子非共生萌发较适宜培养基为1/2MS+1.5mg·L~(-1) 6-BA+0.6mg·L~(-1) NAA+0.75mg·L~(-1) AC+2.5%蔗糖+0.46%琼脂+100mL·L~(-1)椰汁;果实的成熟度影响种子萌发所历经的胚龄、原球茎的颜色及其后续发育,果龄过短或过长都不利于种子发育,带叶兜兰离体培养的最佳时间为果龄180d。

摘  要:
Taking the fruits of Paphiopedilumhirsutissimum from the allogeneic pollination as materials,under the condition of asymbiotic germination,the test had been done of the seed germination of P.hirsutissimumby the orthogonal test,in order to provide the basis of the rapid propagation and industrial production of P.hirsutissimum.The results showed that the asymbiotic germination of P.hirsutissimum was mostly influenced by the basic medium,and the better medium for asymbiotic germination was 1/2MS+1.5mg·L~(-1) 6-BA+0.6mg·L~(-1) NAA+0.75mg·L~(-1) AC+2.5%sucrose+0.46%agar+100mL·L~(-1) coconut juice;the embryo ages,the protocorm color and the protocorm growth were affected by the fruit maturity,the fruit age was harmful to the seed germination,which was too young or too old,and the180-day-old fruit was the optimal in vitro cultivation of P.hirsutissimum.
