橘绿木霉菌丝体中哌珀霉素a、b的提取及测定

摘  要:
[目的]建立发酵液中橘绿木霉(Trichoderma citrinoviride)菌丝体所产生的哌珀霉素a、b 2个代谢产物的提取和含量测定方法。[方法]采用单因素试验和正交试验,对浸提过程中所使用的乙醇浓度、液料比、提取时间和温度等影响因素进行筛选。在使用HPLC测定哌珀霉素a、b的含量时,其测定的色谱条件为Venusil MP C18色谱柱;以乙腈-水(体积比60∶40)为流动相;流速1.0 m L/min;柱温40℃;2个组分的检测波长均为205 nm。[结果]经优化的提取条件为乙醇浓度70%;液料比(v/w)=5∶1;温度和时间:室温下100 min。用HPLC测定哌珀霉素a和b的线性范围分别为10~150、9.1~137 mg/L;平均回收率分别为100.1%(RSD=1.6%)和99.6%(RSD=2.0%);哌珀霉素a、b粗提物的含量分别为232、210 mg/g。[结论]在此研究中所开发的提取和测定方法简便、可靠,能准确测定橘绿木霉菌丝体产生的次级代谢物哌珀霉素a、b的含量。

摘  要:
[Aims] The aims were to establish a methods for extraction and determination of the secondary metabolites of peptaibol a and b from mycelia of Trichoderma citrinoviride during the process of liquid fermentation.[Methods] The single factor and orthogonal test were conducted to screen and optimize the impact factors such as concentration of ethanol,ratio of liquid to solid,extraction time and temperature.The detection of peptaibol a and b was conducted by HPLC used Venusil MP C_(18) column with column temperature of 40 ℃ at 205 nm,in addition,the mixture of acetonitrile and water(60∶40,by vol) was used as the mobile phase with a flow rate of 1.0 m L/min.[Results] The optimized parameters for extraction were: 70% of ethanol,ratio of liquid to solid(v/w) 5:1,room temperature for 100 min.The linear ranges for peptaibol a and b were 10-150 mg/L and 9.1-137 mg/L,the average recoveries were 100.1%(RSD=1.6%)and 99.6%(RSD=2.0%),the contents of the crude peptaibol a and b were 232 and 210 mg/g,respectively.[Conclus ion] The method is simple and reliable,which can accurately detect the contents of peptaibol a and b generated by mycelia of T.citrinoviride.
