A-FABP基因内含子2多态性及A-FABP基因在鹅组织中的差异表达研究

摘  要:
采用PCR-RFLP技术检测160只雄性五龙鹅和172只雄性朗德鹅A-FABP基因内含子2的遗传变异,并与体重和肥肝性状进行关联分析;同时利用Real-timePCR技术检测A-FABP基因在朗德鹅10种组织中的表达情况。结果表明:克隆的五龙鹅和朗德鹅A-FABP基因内含子2序列长461bp(GenBank登录号为DQ647701、EF639390)。该扩增产物酶切后,发现有HaeⅢ-RFLP,并出现AA、AB和BB3种基因型;五龙鹅和朗德鹅的基因型频率分别为0.7、0.2、0.1与0.372、0.465、0.163。填饲的朗德鹅在体重和肥肝重上基因型AA与AB和BB之间存在显著差异(P<0.05),而五龙鹅在体重性状上3种基因型之间差异不显著(P>0.05);A-FABP基因在朗德鹅各组织中的表达量为肝>脂肪>腿肌>脾>肺>心>胸肌>肾>睾丸>脑,A-FABP基因在肝脏和脂肪的表达量之间差异不显著(P>0.05),而与其他组织的表达量相比差异显著(P<0.05)。

摘  要:
The genetic variation of intron 2 of A-FABP gene and is relationship with weight and liver traits in male Wulong Geese and Langde Geese were examined by PCR-RFLP At the same time, using Real-time PCR detected expression of the A-FABP genes in ten tissues of Langde Geese. The results showed that the sequences containing intron 2 of A-FABP gene were 461 bp in length (the GenBank accession number: DQ647701, EF639390). Three genotypes AA, AB and BB and HaeⅢ-RFLP were identified when PCR products were cut by AspⅠ, Hin fⅠand Hae Ⅲ restriction enzyme and Wulong and Langde geese′s genotype frequencies were 0.7, 0.2, 0.1 and 0.372, 0.465, 0.163, respectively. The results of the least squares means showed that genotype AA and AB or BB affected significantly body weight and liver traits in Langde geese (P < 0.05), however, the significant difference was not found in body weight in Wulong geese(P > 0.05). Real-time PCR was used to determine the expression of A-FABP gene in ten tissues of Langde Geese. The result was liver>fat>leg muscle>spleen>lung>heart>chest muscle> kidney>spermary>brain, and the A- FABP expression in liver and fat tissue was not significant (P > 0.05), which was different comparing with the content in other tissues(P < 0.05).
